چکیده

Correction Following publication of the original article [1], the authors were alerted by a colleague of a column duplication in Table 1. Since the summary row was correct, though, the interpretation and the conclusion of the article were not affected. The authors have provided the correct table, and would like to acknowledge the assistance of their colleague in the following words: “We thank Maia Segura-Wang for helpful corrections.”

Optimization and validation of sample preparation for metagenomic sequencing of viruses in clinical samples

BACKGROUND Sequence-specific PCR is the most common approach for virus identification in diagnostic laboratories. However, as specific PCR only detects pre-defined targets, novel virus strains or viruses not included in routine test panels will be missed. Recently, advances in high-throughput sequencing allow for virus-sequence-independent identification of entire virus populations in clinical ...

Preparation of metagenomic libraries from naturally occurring marine viruses.

Microbes are now well recognized as major drivers of the biogeochemical cycling that fuels the Earth, and their viruses (phages) are known to be abundant and important in microbial mortality, horizontal gene transfer, and modulating microbial metabolic output. Investigation of environmental phages has been frustrated by an inability to culture the vast majority of naturally occurring diversity ...

Human Virome

Viruses are dominant entities in the biosphere and parasitize all cellular life forms. The relative abundances of different classes of viruses are dramatically different between prokaryotes and eukaryotes. In marine, soil and animal-associated environments, virus particles consistently outnumber cells by one to two orders of magnitude. It is estimated that 10 quintillion (1030) viral particles ...

Virus detection using metagenomic sequencing of swine nasal and rectal swabs

Advances in DNA sequencing have increased our ability to generate large amounts of sequence data at lower costs. These developments have enabled microbial detection and characterization directly from clinical specimens, known as metagenomic sequencing. Viral metagenomic sequencing was performed on five nasaland five fecal-swab pools collected from each of two primary and two secondary market sl...

Rapid Metagenomic Next-Generation Sequencing during an Investigation of Hospital-Acquired Human Parainfluenza Virus 3 Infections

Metagenomic next-generation sequencing (mNGS) is increasingly used for the unbiased detection of viruses, bacteria, fungi, and eukaryotic parasites in clinical samples. Whole-genome sequencing (WGS) of clinical bacterial isolates has been shown to inform hospital infection prevention practices, but this technology has not been utilized during potential respiratory virus outbreaks. Here, we repo...
